N-Boc-4-piperidine methanol was dissolved in anhydrous chloroform, 10 mL of anhydrous chloropropylamine solution was added, 5 mL of chloroform solution of p-toluenesulfonic acid was added, 4A molecular sieve was added, the reaction mixture was refluxed for 96 h, after the reaction was completed, the mixture was filtered through Na2CO3, the solvent was removed from the filtrate, and the crude product was recrystallized in isopropyl ether to obtain pure tert-butyl 2,9-diazaspiro[5.5]undecane-2-carboxylate.
