以6-溴-4-氟-1-异丙基-2-甲基-1H-苯并[D]咪唑和联硼酸频那醇酯为原料,合成4-氟-1-异丙基-2-甲基-6-硼酸频哪醇酯基-1H-苯并[d]咪唑的一般步骤如下:在500 mL单颈烧瓶中加入DMSO(100 mL),随后依次加入6-溴-4-氟-1-异丙基-2-甲基-1H-苯并[D]咪唑(20.0 g,73.80 mmol)、双(频哪醇合)二硼(27.6 g,108.69 mmol)、三环己基膦(3.53 g,12.61 mmol)和乙酸钾(21.3 g,217.38 mmol)。迅速加入乙酸钯(1.5 g)后,向反应烧瓶中通入氮气进行保护,随后在氮气氛围下加热至90℃。反应3小时后,将反应烧瓶冷却至室温,将反应混合物倒入700 mL水中,充分搅拌后进行减压过滤。滤饼用100 mL水洗涤两次,干燥后得到浅棕色固体。将粗产物用50 mL石油醚和10 mL乙酸乙酯打浆10分钟,经过真空过滤得到白色固体(18.8 g,收率80.3%)。
