| Description | Aviptadil (INN) is a synthetic analog of vasoactive intestinal peptide (VIP), possessing potent vasodilatory effects. Aviptadil induces pulmonary vasodilation, inhibits vascular smooth muscle cell (SMC) proliferation and platelet aggregation. Aviptadil can be used for research on pulmonary fibrosis, pulmonary arterial hypertension (PAH), and respiratory failure caused by SARS-CoV-2. |
| In vitro | Method: Aviptadil (the study used vasoactive intestinal peptide, VIP) was applied to rat alveolar L2 epithelial cells exposed to 0.1–1.75% cigarette-smoke extract for 48 h. Cell death, apoptosis, cAMP generation, MAPK signaling and specific 125I-VIP binding were examined. Result: VIP at concentrations above 10^-13 M concentration-dependently reduced cigarette-smoke-extract-induced apoptotic cell death. The protective response was associated with VIP-receptor signaling, cAMP-dependent protein kinase and MAPK activation.[1] |
| In vivo | Method: Aviptadil (the study used vasoactive intestinal peptide, VIP) was evaluated in Sprague-Dawley rats with monocrotaline-induced pulmonary arterial hypertension. In a prevention protocol, VIP was administered intraperitoneally at 500 μg/kg every other day for 3 weeks, beginning on the day of monocrotaline treatment; ten injections were given. VIP was also studied alone or with bosentan in treatment protocols. Result: Preventive VIP treatment markedly inhibited development of pulmonary hypertension, vascular remodeling and mortality. VIP also improved established disease, while the combination with bosentan generally produced greater effects than either treatment alone.[2] Method: Aviptadil (the study used vasoactive intestinal peptide, VIP) was continuously delivered by an intraperitoneal osmotic pump to adult male Sprague-Dawley rats after creation of an aortocaval shunt. Four weeks later, airway responsiveness was assessed during intravenous methacholine infusion at 2, 4, 8, 12 and 24 μg/kg/min using forced-oscillation measurements. Result: Chronic VIP treatment prevented development of bronchial hyperreactivity associated with the precapillary pulmonary-hypertension model.[3] |
