DescriptionCcpA (Catabolite Control Protein A) is a global transcriptional regulator in Gram-positive bacteria that governs carbon catabolite repression and metabolic flux. It directly binds to catabolite-responsive elements (cre) in target promoters, such as the ackA and pta genes, modulating their expression in response to carbohydrate availability and environmental pH. Research applications include investigating how CcpA coordinates virulence factor expression, biofilm formation (e.g., through synergistic control with CidR of the cidABC operon), and bacterial stress tolerance, including sensitivity to antimicrobial fatty acids. Deletion of ccpA alters infectivity in hyperglycemic hosts and rebalances bacterial growth versus host tissue damage in pathogenesis models.
In vitroCCPA suppressed the mRNA and protein expressions of CnAβ and exerted antihypertrophic effects to a greater degree than CsA. The inhibition of CCPA on cardiomyocyte hypertrophy was counteracted by the A1 receptor antagonist DPCPX[1].
In vivoCCPA (CCPA) was synthesized as a potential high affinity ligand for A1 adenosine receptors. Binding of [3H]PIA to A1 receptors of rat brain membranes was inhibited by CCPA with a Ki-value of 0.4 nM, compared to a Ki-value of 0.8 nM for the parent compound N6-cyclopentyladenosine (CPA)[2].
