| Description | CMP3a is a potent and selective NEK2 kinase inhibitor (IC50 = 15 nM) that leads to EZH2 protein destabilization by inhibiting NEK2 kinase activity. In preclinical models of glioblastoma (GBM), CMP3a inhibits tumor growth and exhibits synergistic effects with radiotherapy. |
| In vitro | Method: CMP3a (the study used compound 3a, CMP3a) was tested in a cell-free NEK2 kinase assay and in patient-derived glioma spheres, including the 528 glioma stem-like cell model.
Result: CMP3a inhibited NEK2 with an IC50 of 82.74 nM, destabilized EZH2, and reduced glioma-sphere proliferation and self-renewal; combination with radiation produced greater effects than either treatment alone.[1] |
| In vivo | Method: CMP3a was evaluated in glioma stem-cell-derived intracranial xenograft models, alone or with radiation.
Result: Pharmacological NEK2 inhibition attenuated intracranial glioblastoma growth and radioresistance. [1] |
