Tacrolimus is a macrolide‑type substance isolated from Streptomyces. It exhibits potent immunosuppressive activity. It specifically binds to and inhibits the activity of calcineurin, suppresses IL‑2 signal transcription, thereby inhibiting T‑cell activation, reducing the production of TNF‑α, IL‑1β and IL‑6, as well as T‑cell‑dependent B‑cell proliferation. Compared with cyclosporin, another calcineurin inhibitor, tacrolimus has stronger efficacy in suppressing T‑cell activation with fewer adverse reactions.
