Applications of Urokinase 

Urokinase (also called urokinase-type plasminogen activator, uPA) is a serine protease that converts plasminogen into plasmin, leading to fibrin clot breakdown (thrombolysis). Its clinical and research applications include:  

1. Therapeutic Uses (Thrombolysis) 

- Acute Myocardial Infarction (AMI): Dissolves coronary artery blood clots (though largely replaced by tissue plasminogen activators like **alteplase**).  

- Pulmonary Embolism (PE): Breaks down clots in pulmonary arteries to restore blood flow.  

- Deep Vein Thrombosis (DVT): Used in severe cases to prevent complications like post-thrombotic syndrome.  

- Peripheral Arterial Occlusion: Treats acute limb ischemia caused by arterial clots.  

- Ischemic Stroke (limited use): Sometimes used in specific cases where tPA (alteplase) is unavailable.  

2. Catheter/Dialysis-Related Clot Clearance  

- Occluded IV Catheters or Hemodialysis Shunts: Instilled locally to restore patency.  

3. Local/Non-Systemic Applications  

- Ophthalmology: Treats retinal vein/artery occlusion.  

- Pleural/Peritoneal Effusions: Helps drain viscous hemothorax, empyema, or ascites by breaking fibrin deposits.  

4. Laboratory & Research Applications 

Cell Culture: Detaches adherent cells (e.g., in tissue dissociation protocols).  

Cancer Research: Studies tumor metastasis (uPA is linked to extracellular matrix degradation).  

Fibrinolysis Studies: Investigates clot dissolution mechanisms.
