| Description | Vantictumab (OMP-18R5) is a humanized anti-FZD1/2/5/7/8 monoclonal antibody that binds to frizzled (FZD) receptors and inhibits Wnt pathway signaling. Vantictumab can be used in research on metastatic HER2-negative breast cancer and metastatic pancreatic adenocarcinoma. |
| In vitro | Method: MCF-7, T47D, 293T, SUM149, SUM159, Wnt-secreting L cells, MDA-MB-231, and MDA-MB-231_BH cells were incubated with vantictumab at a concentration of 50 μg/mL. Cells were cultured for 5 days (cell lines) or 7 days (primary samples), after which mammospheres with diameters greater than 50 μm were counted.
Result: Vantictumab inhibited Wnt signaling and reversed CM-induced mammosphere formation in early breast cancer samples in vitro. [1] |
| In vivo | Method: For subcutaneous xenograft experiments, 1 × 10⁶ MCF-7 cells were injected into each flank of female NSG mice. Mice were treated with vantictumab (15 mg/kg, intraperitoneal injection, once weekly) or control vehicle (sterile PBS), starting 3 days before tumor cell injection and continuing throughout the study period.
Result: Vantictumab-treated mice exhibited a significant reduction in ex vivo mammosphere formation by cells isolated from the femur compared with control mice. [1]
Method: A total of 5 × 10⁵ GFP- and luciferase-labeled MDA-MB-231_BH cells were injected intravenously via the tail vein into female BALB/c nude mice. Mice received intraperitoneal injections of vantictumab at a dose of 15 mg/kg once weekly.
Result: Vantictumab reduced spontaneous hindlimb bone metastasis in the MDA-MB-231_BH model. [1] |
