背景信息

Tuberin, encoded by the *TSC2* gene, is a critical tumor suppressor protein that forms a complex with Hamartin (TSC1) to regulate cell growth and metabolism. This TSC1-TSC2 complex inhibits the mechanistic target of rapamycin (mTOR) signaling pathway by acting as a GTPase-activating protein (GAP) for Rheb (Ras homolog enriched in brain), thereby suppressing mTORC1-mediated anabolic processes.
