ethyl 3-methoxy-2-oxo-l,2-dihydropyridine-4-carboxylate [00194] To a solution of ethyl 3-hydroxy-2-oxo-l,2-dihydropyridine-4-carboxylate (Intermediate IB) (500 mg, 2.73 mmol) in acetonitrile (17 mL) and ethyl ether (1.9 mL) was cooled to 0 °C and trimethylsilyldiazomethane (1.50 mL, 3.00 mmol) was added. The reaction mixture was stirred at rt for 30 h. The reaction mixture was filtered and the filtrate was evaporated under reduced pressure to provide the crude product. The crude product was purified via ISCO flash chromatography on silica gel using a 15 min gradient from 0 to 100% EtOAc in hexane to give Example 33 A (330 mg, 1.67 mmol, 61.3 % yield).
