以化合物(CAS: 188290-36-0)为原料合成2,5-二溴噻吩的一般步骤如下:参照文献方法(Kellogg RM, Schaap AP, Harper ET, Wynberg NDH. Acid-catalyzed bromination, deuteration, rearrangement, and debromination of thiophenes under mild conditions. J Org Chem 1968; 33: 2902-9)进行制备。向噻吩(14.0 g, 0.16 mol)溶于250 mL 50:50(v/v)乙酸和氯仿混合溶剂的溶液中,加入N-溴代琥珀酰亚胺(60 g, 0.33 mol)。将反应混合物回流15分钟。反应完成后,冷却至室温,加入水,用氯仿进行萃取。合并有机相,用无水硫酸钠干燥,减压浓缩除去溶剂。粗产物通过柱色谱法纯化,以己烷为洗脱剂,得到目标化合物2,5-二溴噻吩,为液体(31.5 g, 产率84%)。
