Dihydrodutasteride Usage And Synthesis
UsesDihydro Dutasteride is an impurity arose during the process development of Dutasteride (D735000).
Synthesis16.65 g of methyl 3-keto-4-aza-5-androsta-17-carboxylate was dissolved in 250 ml of dichloromethane and 18.95 ml of boron tribromide was added dropwise under cooling in room temperature water (25 C). Stirring was continued at room temperature for 20 min to obtain a clear brown solution. 19.5 ml of 2,5-bis(trifluoromethyl)aniline was added to the above solution under room temperature water (25 C) cooling. The reaction was heated to 50C with stirring overnight. After the reaction solution was cooled to room temperature, 50 mL of water was slowly added and the resulting hydrogen bromide gas was introduced into the aqueous Na2CO3 solution. Continue to add 200 ml of water until all solids are dissolved and a clear two phase is obtained. The lower dichloromethane organic phase was separated and washed with 200 ml of water and 200 ml of saturated saline and dried over anhydrous Na2SO4. It was filtered through 10 g silica gel and concentrated under reduced pressure to 60 mL. Slowly add 100 ml of petroleum ether dropwise to the above concentrate, the product precipitates as a yellowish solid dutasteride dihydrogen, which is filtered, washed with petroleum ether and dried in air to a constant weight: 24.65 g (93%).
