| Preparation | Dissolve 20.4 g (2.0 eq) of disodium hydrogen phosphate in 100 mL of water. After the system is clear, add 160 mL of dichloromethane and N-[(S)-1-ethoxycarbonyl-3-phenylpropyl]-L-alanine (20.0 g, 1.0 eq), and stir at room temperature. Dissolve 8.4 g (0.4 eq) of triphosgene in 40 mL of dichloromethane and add it dropwise to the reaction system at room temperature. After the addition is complete, react for 1 h, then add 0.5 mL of pyridine, stir for 1 h, and allow to stand. Separate the liquid and concentrate the organic phase under reduced pressure to obtain an oily substance, dissolve it in 16 mL of dichloromethane, and add it dropwise to 200 mL of n-heptane. Stir at 0–10 °C for 1 h to induce crystallization. Filter under reduced pressure to obtain a white solid N-[(S)-1-ethoxycarbonyl-3-phenylpropyl]-L-alanine-N-carboxylic anhydride, and dry under vacuum at 50 °C for 10 h. 20.4 g of the product is collected, with a yield of 91.6%. Add L-proline (9.8 g, 1.3 eq), sodium hydroxide (3.4 g, 1.3 eq), 100 mL of water, and 70 mL of tetrahydrofuran to a 250 mL three-necked flask and stir to dissolve at room temperature. Cool the flask to 0–10 °C in an ice-water bath, and dropwise add a solution of N-[(S)-1-ethoxycarbonyl-3-phenylpropyl]-L-alanine-N-carboxylic anhydride (20.0 g, 1.0 eq) in 70 mL of tetrahydrofuran. After the addition is complete, maintain the temperature at 0–10 °C and continue stirring for 2 hours. Adjust the pH to 6 with hydrochloric acid, and evaporate the tetrahydrofuran under reduced pressure. The resulting aqueous solution was adjusted to pH 4.2 with hydrochloric acid, extracted with ethyl acetate (200 mL × 4), and the resulting oily substance was concentrated under reduced pressure. The solution was dissolved in 40 mL of ethyl acetate, and a 180 mL ethyl acetate solution of maleic acid (7.6 g, 1.0 eq) was added dropwise under heating at 50 °C. After the addition was complete, the reaction was continued at this temperature for 1 h, then naturally cooled to 20–30 °C, and crystallization was allowed to continue for 1 h. The product was then filtered under reduced pressure to obtain a white solid, crude enalapril maleate. The solid was washed with 40 mL of ethyl acetate, and the sample was dried under vacuum at 50 °C for 10 h. Yield: 26.1 g, 81.7%. Add 26.0 g of crude enalapril maleate and 200 mL of water to a 500 mL three-necked flask. Start stirring and heat to 65 °C. After the system is completely dissolved, continue heating for 30 min. Turn off the heating and allow it to cool naturally to room temperature. A white solid precipitates. Continue stirring in an ice-water bath (0–10 °C) for 2 h. Filter under reduced pressure. Wash the filter cake with 50 mL of water. Dry the sample under vacuum at 50 °C for 10 h to obtain 20.9 g of white solid. Yield: 80.6%. hydrogenated at 25°C and at 40 psig of hydrogen until uptake of hydrogen
ceases. The solids are filtered, washed with 80 ml of ethanol and the filtrates
are combined. Assay by high pressure liquid chromatography shows an 87:13
ratio of diastereoisomers in favor of the desired product. Ethanol is removed
under vacuum to afford an oil which is dissolved in 60 ml of water and 20 ml
of ethyl acetate. The pH of the stirred two-phase mixture is adjusted to 8.6
with 50% NaOH. The layers are separated and the water phase is extracted
with 2x20 ml of ethyl acetate. The water phase is adjusted to pH 4.25 with
hydrochloric acid, 12 g of NaCl is dissolved in the water, and product is
extracted with 5x12 ml of ethyl acetate. The extracts are combined and dried
with Na2SO4. The desired product, N-[1-(S)-ethoxycarbonyl-3-phenylpropyl]-
L-alanyl-L-proline, is crystallized as its maleate salt by addition of 1.86 g of
maleic acid. After stirring for 4 hours, the salt is filtered, washed with ethyl
