5-Bromo-4-chloro-3-indolyl-β-D-glucoside (X-Gluc) is a chromogenic substrate widely used in molecular biology and biochemistry to detect β-glucosidase enzyme activity.

Mechanism: When cleaved by β-glucosidase (or the related enzyme β-glucuronidase, depending on derivative), it releases an insoluble blue indigo dye (5,5′-dibromo-4,4′-dichloro-indigo).

Note: While X-Gluc typically refers to the β-D-glucopyranoside, a closely related compound—X-GlucA (5-bromo-4-chloro-3-indolyl-β-D-glucuronide)—is more commonly used for GUS (β-glucuronidase) assays in plant molecular biology.
