Descriptionm-PEG8-NHS ester is a PEG linker containing an NHS ester. The NHS ester can be used to label the primary amines (-NH2) of proteins, amine-modified oligonucleotides, and other amine-containing molecules. The hydrophilic PEG spacer increases solubility in aqueous media.
Usesm-PEG8-NHS ester is a non-cleavable 8 unit PEG ADC linker used in the synthesis of antibody-drug conjugates (ADCs).
Applicationm-PEG8-NHS ester (CAS 756525-90-3) is a high-purity, heterobifunctional crosslinker. It is characterised by a methoxy-terminated polyethylene glycol (PEG) chain comprising eight repeating units, and an N-hydroxysuccinimide (NHS) ester group capable of reacting with amines. The PEG linker confers hydrophilicity to the molecule, thereby enhancing the solubility of the resulting bioconjugate and reducing its immunogenicity. The NHS ester facilitates covalent bonding with primary amines on biomolecules such as proteins, peptides and amine-modified oligonucleotides; the optimal pH for this reaction is 7.2–8.5.

storagem-PEG8-NHS ester should normally be stored for long periods in tightly sealed vials at –20 °C, in a dry place, protected from light and under a nitrogen or argon atmosphere. It is important to note that, after removal from the refrigerator, the vial must be left to stand in a dry place until it has returned to room temperature before opening; otherwise, moisture in the air will instantly condense inside the vial, causing the NHS ester to hydrolyse into a carboxylic acid and rendering it completely ineffective.
